目的:研究大鼠单侧睾丸损伤后对侧睾丸组织变化及其发生机制。方法:选择SD雄性大鼠40只,随机分为4组:A组为对照组,B组为单侧睾丸挫伤组,C组为单侧睾丸裂伤组,D组为裂伤后腹腔注射环孢素A(cyclosporin A,CsA)组。分别于第2、4周观察大鼠后足垫反应(foot pad reaction,FPR),取对侧睾丸做苏木精-伊红(HE)染色和免疫组化检测CD_4分子、CD_8分子的表达。结果:注射睾丸抗原48 h后,B、C组大鼠右足足垫较A、D组显著增厚(P<0.01),C组与B组差异亦有显著统计学意义(P<0.05),A组与D组差异无统计学意义(P>0.05)。B、C组对侧睾丸组织学发生明显改变,A、D组改变不明显。B、C组中CD_4~+T细胞平均光密度(OD)值高于CD_8~+T细胞(P<0.05),两种T细胞OD值均明显高于对照组(P<0.01),C组与B组比较,CD_4~+T细胞OD值差异有统计学意义(P<0.05),CD_8~+T细胞OD值差异无统计学意义(P>0.05)。各组结果在第2、4周比较,差异均无统计学意义。结论:单侧睾丸损伤可引起对侧睾丸组织发生迟发型超敏反应(delayed type hypersensitivity,DTH),主要由CD_4~+T细胞介导,且与睾丸损伤程度有关。CsA可抑制DTH的发生。
Objective: To investigate the changes of contralateral testicular tissue after unilateral testicular injury in rats and its mechanism. Methods: Forty male Sprague-Dawley rats were randomly divided into 4 groups: group A as control group, group B as unilateral testicular contusion group, group C as unilateral testicular laceration group, and group D as laceration after retroperitoneal injection Cyclosporin A (CsA) group. Fetal footpad reaction (FPR) was observed in the 2nd and 4th week respectively. Hematoxylin and eosin (HE) staining and immunohistochemistry were used to detect the expression of CD_4 and CD_8 in the contralateral testes. RESULTS: After injection of testis antigen for 48 h, the right foot footpad of rats in group B and C were significantly thicker than those in group A and D (P <0.01), and the difference between group C and B was also statistically significant (P <0.05) There was no significant difference between group A and group D (P> 0.05). B, C group contralateral testicular histology significantly changed, A, D group did not change significantly. The OD value of CD_4 ~ + T cells in group B and group C were higher than those in CD_8 ~ + T cells (P <0.05), OD values of both T cells were significantly higher than those in control group (P <0.01) Compared with group B, the OD value of CD_4 ~ + T cells was significantly different (P <0.05). There was no significant difference in OD value between CD_8 ~ + T cells (P> 0.05). The results of the two groups in the first two weeks, the difference was not statistically significant. CONCLUSION: Unilateral testicular injury can cause delayed type hypersensitivity (DTH) in the contralateral testicular tissue, which is mainly mediated by CD_4 ~ + T cells and is related to the degree of testicular injury. CsA can inhibit the occurrence of DTH.
