用物理化学方法纯化的人血浆蛋白C(HPC)免疫小鼠,制备了8株分泌抗HPC单克隆抗体的杂交瘤细胞株,分别命名为PC01~PC08。各株杂交瘤细胞都具有长期稳定的抗体分泌能力,其中PC03为构象特异性单克隆抗体,二价金属离子对其与HPC的结合有明显影响;PC02和PC03对活性蛋白C的酶活性有促进作用;而PC04具有抑制作用。
Eight kinds of hybridoma cell lines secreting anti-HPC McAbs were prepared by immunizing mice with the physicochemical method of purified human plasma protein C (HPC), and named PC01 ~ PC08 respectively. Each hybridoma cell has long-term stable antibody secretion ability, PC03 is a conformation-specific monoclonal antibody, and bivalent metal ion has a significant effect on its binding with HPC; PC02 and PC03 promote the activity of active protein C Role; while PC04 has an inhibitory effect.
