PCR法扩增人甲状腺过氧化物酶(hTPO)抗原决定簇基因(1702~2622)片段并插入到载体之中,获得2个参与自身免疫反应的B细胞抗原决定簇C2和C21、一个T细胞抗原决定簇和一个可能的B细胞抗原决定簇。用ApaI和SacI双酶切此hTPO基因片段可获得3个较短的肽段,行酶切鉴定后再分别亚克隆,从而为今后的序列测定和hTPO表达产物及其单克隆抗体的研究提供目的基因。
PCR amplification of human thyroid peroxidase (hTPO) epitopes gene (1702 ~ 2622) fragment and inserted into the vector to obtain two autoimmune responses of B cell epitopes C2 and C21, a T cell Antigenic determinant and a possible B-cell epitope. Three digests of the hTPO gene fragment were digested with ApaI and SacI to obtain three shorter peptides, which were subcloned separately after digestion with restriction enzyme digestion to provide the objective for future sequencing and study of hTPO expression products and their monoclonal antibodies gene.
