目的 评价电化学疗法 (EChT)对大鼠种植型肝癌的治疗效果。材料与方法 采用直接注射法制作大鼠肝癌模型。应用影像学、病理学及系列免疫指标 (IL 2、sIL 2R、IL 6、TNF α)检测 ,生存期比较等方法观察荷瘤大鼠EChT治疗前后肿瘤大小、坏死、细胞凋亡、外周血系列免疫指标表达水平、生存期长短的动态变化 ,并与同期对照组进行比较。结果 EChT治疗前 ,大鼠肝肿瘤平均体积为 10 0± 6mm3 ;EChT治疗后 1周 ,大鼠肝肿瘤平均体积为12 5± 10mm3 ,而荷瘤对照组肝肿瘤平均体积为 190± 11mm3 ;治疗组肿瘤生长率为 2 5 % ,而荷瘤对照组肝肿瘤生长率为 90 % (P <0 .0 5 )。治疗组病理光镜下肿瘤明显坏死 ,并可明显诱发细胞凋亡。荷瘤大鼠EChT治疗前IL 2水平呈低表达 ,sIL 2R、IL 6、TNF α则高表达 ,与正常大鼠比较差异显著 (P <0 .0 5 ) ;EChT治疗后 ,与同期荷瘤大鼠对照组比较 ,IL 2表达水平升高 ,sIL 2R、IL 6、TNF α表达水平下降 (P <0 .0 5 )。EChT治疗组大鼠生存期明显延长 (P <0 .0 5 )。结论 肿瘤负荷可导致大鼠细胞因子表达紊乱。EChT治疗可明显抑制肿瘤生长 ,促进肿瘤坏死 ,诱发肿瘤细胞凋亡 ,改善和纠正机体免疫紊乱 ,增强大鼠免疫力 ,延长大鼠生存期。
Objective To evaluate the therapeutic effect of electrochemical therapy (EChT) on implanted hepatocellular carcinoma in rats. Materials and Methods The model of rat liver cancer was made by direct injection. The tumor size, necrosis and apoptosis were observed before and after EChT treatment in tumor-bearing rats by imaging, pathology and serial immune indicators (IL 2, sIL 2R, IL 6, TNFα) Immune indicators expression level, the dynamic changes of the length of life, and compared with the same period the control group. Results Before EChT treatment, the average volume of rat liver tumors was 100 ± 6mm3. One week after EChT treatment, the average volume of rat liver tumors was 125 ± 10mm3, while the average tumor volume of tumor-bearing control tumors was 190 ± 11mm3. Group tumor growth rate of 25%, while the tumor-bearing control group, the growth rate of liver tumors was 90% (P <0.05). In the treatment group, the tumor was obviously necrotic under light microscope, and apoptosis was obviously induced. The level of IL-2 in tumor-bearing rats before EChT treatment was low, and the levels of sIL 2R, IL-6 and TNF-α were higher than those in normal rats (P <0.05). After EChT treatment, Compared with the control group, IL 2 expression increased and sIL 2R, IL 6 and TNFα levels decreased (P <0.05). The survival of rats in EChT treatment group was significantly prolonged (P <0.05). Conclusion Tumor burden can lead to disorder of cytokine expression in rats. EChT treatment can significantly inhibit tumor growth, promote tumor necrosis, induce apoptosis of tumor cells, improve and correct immune disorders, enhance immunity and prolong the survival of rats.
