目的研究壳寡糖对雌二醇诱导的人乳腺癌MCF-7细胞分泌促肿瘤血管生成因子的抑制作用。方法采用MTT法检测处理24和48 h后壳寡糖对正常乳腺癌MCF-7细胞生长的影响;采用RT-PCR方法对MCF-7分泌促肿瘤血管生成因子MMP-9,TIMP-1,2和VEGF在mRNA水平上的变化进行考察。结果壳寡糖对没有经过雌二醇诱导的人乳腺癌MCF-7细胞的生长没有明显作用;雌二醇能够显著上调MCF-7细胞MMP-9和VEGF的mRNA水平;50μg·mL~1的壳寡糖能够显著下调由雌二醇诱导的MMP-9的mRNA水平,但是对TIMP-1,2和VEGF则没有明显作用。结论壳寡糖对雌二醇诱导的乳腺癌MCF-7细胞产生促肿瘤血管生成因子MMP-9在mRNA水平上具有显著抑制作用。
Objective To study the inhibitory effect of chitooligosaccharides on estradiol-induced tumor angiogenesis factor secretion in human breast cancer MCF-7 cells. Methods MTT assay was used to detect the effect of chitooligosaccharides on the growth of MCF-7 breast cancer cells at 24 and 48 h after treatment. The expressions of tumor angiogenesis factors MMP-9, TIMP-1, 2 And VEGF in the mRNA level changes were investigated. Results Chitooligosaccharides had no significant effect on the growth of human breast cancer MCF-7 cells induced by estradiol. Estradiol significantly increased the mRNA and protein levels of MMP-9 and VEGF in MCF-7 cells. Chitooligosaccharides significantly down-regulated the mRNA level of MMP-9 induced by estradiol but had no significant effect on TIMP-1,2 and VEGF. Conclusion Chitooligosaccharides have a significant inhibitory effect on the mRNA expression of pro-angiogenic factor MMP-9 in estradiol-induced breast cancer MCF-7 cells.
