本文用反相高效液相色谱法测定病人血浆中环丙氟哌酸浓度。以甲醇-乙腈-0.01 mol/L磷酸盐(25: 20:55)为流动相,荧光338/455nm检测。线性范围0.15~5.0μg/ml,平均回收率98.75%~104.32%,日内日间差异<8.18%。以本法测定10例呼吸道感染患者环丙氟哌酸血药浓度并分析药代动力学,结果单剂量口服500 mg环内氟哌酸后,药时曲线呈一级吸收二空模型。T_(max)=1.186±0.472h,C_(max)=2.573±1.198μg/ml。AUC=13.602±6.630μg/ml~(-1)·h,V/F=153.110±66.571L。T_(1/2)β=5.67±1.85h。
In this paper, the concentration of ciprofloxacin in plasma of patients was determined by RP-HPLC. Methanol - acetonitrile - 0.01 mol / L phosphate (25:20:55) as the mobile phase, fluorescence detection 338 / 455nm. The linear range of 0.15 ~ 5.0μg / ml, the average recovery was 98.75% ~ 104.32%, intraday difference between days <8.18%. The method was used to determine the plasma concentration of ciprofloxacin in 10 patients with respiratory tract infection and to analyze the pharmacokinetics. The results showed that the drug-time curve showed a first-order model of absorption after a single oral dose of 500 mg norfloxacin. T max = 1.186 ± 0.472 h and C max = 2.573 ± 1.198 μg / ml.
