目的研究北五味子Schisandra chinensis(Turcz.)Baill.叶的化学成分及化合物五味子醇甲对脂多糖(LPS)诱导的小胶质细胞激活时所分泌的促炎性因子的影响。方法用色谱法分离并通过波谱分析对化合物的结构进行鉴定;采用LPS诱导的小胶质细胞活化模型,以Griess法检测化合物对LPS激活小胶质细胞的抑制作用;以MTT法检测化合物对小胶质细胞细胞成活率的影响。结果从北五味子叶中分离鉴定了8个化合物:β-谷甾醇(1)、五味子醇乙(2)、五味子乙素(3)、五味子丙素(4)、五味子酯乙(5)、五味子醇甲(6)、(-)-戈米辛M1(7)及(-)-戈米辛L1(8)。五味子醇甲可显著抑制激活的N9小胶质细胞的NO释放。结论首次系统分离并鉴定北五味子叶的化学成分,其中五味子醇甲能明显抑制小胶质细胞的活化。
Aim To study the chemical constituents of Schisandra chinensis (Turcz.) Baill. And the effect of schizandrin on secretion of proinflammatory cytokines by lipopolysaccharide (LPS) -induced microglial activation. Methods The compounds were isolated by chromatography and identified by spectroscopic analysis. The microglial activation model induced by LPS was used to detect the inhibitory effect of compounds on LPS-activated microglial cells by Griess method. Effect of glial cell survival rate. Results Eight compounds were isolated and identified from the leaves of Schisandra chinensis: β-sitosterol (1), Schizandrol B (2), Schisandrin B (3), Schisandra B (4) Alcohol A (6), (-) - Gomisin M1 (7) and (-) - Gomisin L1 (8). Schizandrol A significantly inhibited NO release from activated N9 microglial cells. Conclusion For the first time, the chemical constituents of Schisandra sphenanthera were systematically isolated and identified. Schizandrin could significantly inhibit the activation of microglial cells.
