目的评价盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊(恩尔欣)、奥司他韦胶囊(达菲)对甲型流感病毒(H1N1)的抑制效率,检验盐酸阿比朵尔胶囊替代奥司他韦胶囊用于流感治疗的可能性。方法本实验以奥司他韦胶囊为阳性对照药物,采用细胞病变效应(CPE)抑制法、噻唑蓝(MTT)比色法和血凝滴度法对其抗甲型H1N1流感病毒的作用进行了观察。结果盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊、奥司他韦胶囊在安全浓度范围内,对100 TC ID50(病毒半数组织感染量)的甲型H1N1流感病毒均有一定抑制作用,且呈明显量-效关系。其中盐酸阿比朵尔原料药抗病毒生物合成作用、对病毒直接杀伤作用和抗病毒吸附、穿入作用三种方式的治疗指数(TI)分别为(3.08±0.34)、(3.55±0.54)、(3.26±0.32);盐酸阿比朵尔胶囊的分别为(3.12±0.40)、(3.41±0.23)、(4.20±0.43);奥司他韦胶囊的依次为(5.34±0.26)、(1.19±0.16)、(1.01±0.38)。结论盐酸阿比朵尔原料药、盐酸阿比朵尔胶囊在阻断流感病毒吸附、穿入作用和直接杀伤作用方面较奥司他韦胶囊效果好(P<0.01)。奥司他韦胶囊的抗病毒生物合成作用较盐酸阿比朵尔和盐酸阿比朵尔胶囊强(P<0.01),因此盐酸阿比朵尔胶囊可以替代奥司他韦胶囊用于预防甲型H1N1流感病毒感染。
OBJECTIVE: To evaluate the inhibitory effect of abidol hydrochloride API, abiraterone hydrochloride (NEXEX) and oseltamivir (Tamiflu) on the influenza A virus (H1N1) Alternatives to oseltamivir capsules for flu treatment. Methods In this study, oseltamivir capsules as a positive control drug, the use of cytopathic effect (CPE) inhibition, thiazolyl blue (MTT) colorimetric and hemagglutination titer of its anti-influenza A H1N1 influenza virus Observed. Results Abidophor hydrochloride hydrochloride, abiraterol hydrochloride, and oseltamivir both inhibited the influenza A (H1N1) virus with a titer of 100 TC ID50 (half of the virus infection) in a safe concentration range. And showed a significant amount - effect relationship. Among them, the therapeutic index (TI) of abiraterone hydrochloride raw material for antiviral biosynthesis, direct killing of the virus and antiviral adsorption and penetration were (3.08 ± 0.34), (3.55 ± 0.54), (3.26 ± 0.32) and (3.12 ± 0.40), (4.20 ± 0.43) and (5.34 ± 0.26) and (1.19 ± 0.16), (1.01 ± 0.38). Conclusion Arbidol hydrochloride and Abiraterone hydrochloride capsules have better effects than oseltamivir capsules in blocking the adsorption, penetration and direct killing of influenza virus (P <0.01). The effect of antiviral biosynthesis of oseltamivir capsules was stronger than that of abiraterone hydrochloride and abiraterone hydrochloride capsules (P <0.01). Therefore, abiraterone hydrochloride capsules can be used as an alternative to oseltamivir capsules for preventing type A H1N1 influenza virus infection.
