人精液中含少量前列腺素,其中E_1、E_2可增强精子运动力和穿卵能力。为了研究适合阴道用的避孕新药,对前列腺素受体拮抗剂Di-4-根皮素磷酸盐(DPP)抗人精子运动潜力进行了评价。禁欲3~5天,手淫取精,室温下(30℃)液化15~25分钟,精液量超过1.5ml,精子密度大于40×10~6/ml,活动率大于50%,正常形态精子大于60%。在标本中加入8ml BWW缓冲液,经500g离心5分钟3次分离精子,再悬浮至浓度20×10~6/ml,待用。
Human semen contains a small amount of prostaglandin, which E_1, E_2 can enhance sperm motility and ability to wear eggs. In order to study contraceptives suitable for vaginal use, prostaglandin receptor antagonist Di-4-phloretin (DPP) was evaluated for anti-human sperm motility. Abstinence 3 to 5 days, masturbation fine, liquefied at room temperature (30 ℃) for 15 to 25 minutes, the amount of semen more than 1.5ml, sperm density greater than 40 × 10 ~ 6 / ml, activity rate greater than 50%, normal morphology is greater than 60 %. Add 8ml of BWW buffer to the specimen, centrifuge for 5 minutes at 500g for 3 times to separate the sperm and re-suspend it to the concentration of 20 × 10 ~ 6 / ml for later use.
