目的:观察survivin反义寡核苷酸(Antisense Oligodeoxynuleotide,ASDON)对人胆管癌QBC939细胞株凋亡增殖及survivin和cyclin D1表达的影响。方法:合成特异性靶向survivin ASODN,并将其转染QBC939细胞,流式细胞术观察survivin ASODN对QBC939细胞凋亡和增殖的影响;同时,应用免疫组化检测survivin ASODN对QBC939细胞中survivin和细胞周期素D1(cyclin D1)蛋白表达的影响。结果:ASODN转染组细胞凋亡指数明显高于对照组(P<0.05),ASODN转染组细胞增殖指数明显低于各对照组(P<0.05),ASODN组survivin和cyclin D1的蛋白表达明显低于对照组(P<0.05)。结论:Survivin ASODN转染能诱导胆管癌细胞QBC939凋亡,抑制细胞增殖,并可下调survivin和cyclin D1蛋白表达。
OBJECTIVE: To observe the effects of survivin antisense oligonucleotide (ASDON) on apoptosis, proliferation and the expression of survivin and cyclin D1 in human cholangiocarcinoma QBC939 cell line. Methods: Survivin ASODN was specifically targeted and transfected into QBC939 cells. Flow cytometry was used to observe the effect of survivin ASODN on the apoptosis and proliferation of QBC939 cells. Meanwhile, immunohistochemistry was used to detect the expression of survivin and Effect of cyclin D1 protein expression. Results: The apoptosis index of ASODN transfection group was significantly higher than that of control group (P <0.05). The proliferation index of ASODN transfection group was significantly lower than that of control group (P <0.05). The protein expression of survivin and cyclin D1 in ASODN group was significantly Lower than the control group (P <0.05). CONCLUSIONS: Survivin ASODN transfection can induce apoptosis of human cholangiocarcinoma QBC939, inhibit cell proliferation, and down-regulate the expression of survivin and cyclin D1.
