目的:研究中医补肾活血法指导下中药复方(益智健脑方浓缩液)以及针刺(“补肾活血针刺法”)对阿尔茨海默病模型小鼠SAMP8(Senescence Accelerated MouseP8,SAMP8)的行为学影响,从不同侧面阐明补肾活血法对SAMP8小鼠的治疗作用以及防治AD的可行性。方法:将5月龄雄性SAMP830只随机分为中药治疗组(n=10)、针刺治疗组(n=10)和模型对照组(n=10),中药治疗组以益智健脑方浓缩液灌胃;针刺治疗组予以“补肾活血针刺法”干预;模型对照组不予任何处理;另设5月龄雄性SAMR1小鼠(n=10)作为正常对照组。4周后,采用Morris水迷宫对各组小鼠的行为学进行检测分析其学习记忆能力的变化。结果:Morris水迷宫实验中,模型对照组SAMP8小鼠的逃避潜伏期较正常对照组SAMR1小鼠的逃避潜伏期时间明显延长(P<0.05),停留在原平台象限的时间和穿越原平台次数较正常对照组SAMR1小鼠明显减少(P<0.05);补肾活血法指导下,采用中药复方益智健脑方浓缩液以及“补肾活血针刺法”干预均能够明显缩短SAMP8小鼠的逃避潜伏期(P<0.05),增加其停留在原平台象限的时间和穿越原平台次数(P<0.05)。结论:补肾活血法能够明显改善AD模型小鼠SAMP8的学习记忆能力以及记忆巩固、再现能力。
OBJECTIVE: To study the effect of traditional Chinese medicine compound prescription (Yizhi Jiannao Concentrate) and acupuncture (“Kidney-Nourishing and Huoxue Acupuncture”) on SAMP8 in mice with Alzheimer’s disease (SAMP8) The ethical influence of the method is to clarify the therapeutic effect of Bushen Huoxue method on SAMP8 mice and the feasibility of prevention and treatment of AD. METHODS: Five-month-old male SAMP830 were randomly divided into Chinese medicine treatment group (n=10), acupuncture treatment group (n=10) and model control group (n=10). The Chinese medicine treatment group was concentrated in Zhijinnaofang group. The rats in the acupuncture group were treated with “Kidney-nourishing and blood-activating acupuncture method” intervention. The model control group was not treated. The 5-month-old male SAMR1 mice (n=10) were used as the normal control group. After 4 weeks, Morris water maze was used to test the behavior of each group of mice for the change of learning and memory abilities. RESULTS: In the Morris water maze test, the escape latency of the model control group SAMP8 mice was significantly longer than that of the normal control group SAMR1 mice (P<0.05). The time spent in the original platform quadrant and the number of crossings of the original platform were significantly higher than those of the normal control group. The number of SAMR1 mice was significantly decreased (P<0.05); under the guidance of Bushen Huoxue method, the use of Chinese herbal compound Yizhi Jiannao Concentrate and “Kidney-nourishing and activating acupuncture method” intervention can significantly shorten the escape latency of SAMP8 mice ( P<0.05), increasing the time of staying in the quadrant of the original platform and the number of crossings of the original platform (P<0.05). Conclusion: Bushenhuoxue can significantly improve the ability of learning and memory of SAMP8 mice as well as memory consolidation and reproducibility.
