目的研究中草药半边旗5F对系统性硬皮病患者皮肤成纤维细胞的作用,以期找到治疗该病的新的有效药物。方法用四甲基偶氮唑盐(MTT)方法和光度检测法及逆转录-聚合酶链反应(RT-PCR)法测定半边旗5F对成纤维细胞增殖和培养基中的胶原含量以及Ⅰ、Ⅲ型前胶原mRNA表达的影响。结果半边旗5F对患者皮肤成纤维细胞的增殖具有显著的抑制作用,还能显著降低该细胞培养基中可溶性胶原的含量(P<0.01)。上述抑制作用随着药物浓度的增高和作用时间的延长而增强。采用RT-PCR法对Ⅰ、Ⅲ型前胶原的mRNA检测显示随着药物浓度的增高二者的表达量降低。结论半边旗5F对系统性硬皮病患者皮肤成纤维细胞的增殖和胶原合成具有显著的抑制作用,为寻找有效治疗该病的药物提供了新的启示。
Objective To study the effect of the Chinese herbal medicine Pteris 5F on the skin fibroblasts of patients with systemic scleroderma in order to find a new and effective drug for the treatment of this disease. Methods MTT assay and reverse transcription-polymerase chain reaction (RT-PCR) were used to determine the effect of 5F on the proliferation of fibroblasts and the content of collagen in culture medium Ⅰ, Ⅲ procollagen mRNA expression. Results Pteris 5F had a significant inhibitory effect on the proliferation of human skin fibroblasts, but also significantly reduced the content of soluble collagen (P <0.01). The above-mentioned inhibitory effect increases with the increase of the drug concentration and the prolongation of the action time. The mRNA expression of procollagen type Ⅰ and Ⅲ was detected by RT-PCR. The mRNA expression of procollagen Ⅰ and Ⅲ was decreased with the increase of drug concentration. Conclusions Pteris 5F can significantly inhibit the proliferation and collagen synthesis of human dermal fibroblasts in patients with systemic scleroderma, and provide a new inspiration for finding effective drugs for treating this disease.
