目的:建立一种用异硫氰酸苯酯柱前衍生的高效液相色谱方法测定金银花中17种氨基酸含量的方法,为金银花药性物质基础研究提供实验依据。方法:水解金银花总蛋白,以异硫氰酸苯酯进行柱前衍生,用HPLC测定金银花中氨基酸峰面积并计算其含量。结果:各种氨基酸均能达到基线分离。结论:高效液相色谱柱前衍生法可作为金银花药材中初生成分的测定手段之一。
OBJECTIVE: To establish a method for the determination of 17 kinds of amino acids in honeysuckle by high performance liquid chromatography (HPLC) with phenylisothiocyanate pre-column derivatization, and to provide an experimental basis for the basic research of the drug. Methods: The total protein of honeysuckle was hydrolyzed and pre-column derivatization was carried out with phenyl isothiocyanate. The peak area of amino acids in honeysuckle was determined by HPLC and its content was calculated. Results: Various amino acids were able to achieve baseline separation. Conclusion: HPLC pre-column derivatization method can be used as one of the primary constituents of honeysuckle herbs.
