[目的]采用反相高效液相色谱法分析嘧肽霉素发酵液,外标法测定其中抗真菌组分的含量。[方法]使用DIKMA DiamonsilⅡC18反相柱和紫外可变波长检测器,以甲醇-水(体积比为20∶80)为流动相,流速为1 mL/min,检测波长为278 nm。[结果]嘧肽霉素抗真菌组分保留时间为10.2 min,方法的线性相关系数为0.999 8,样品平均回收率为99.4%,变异系数为0.25%。[结论]该方法是用于发酵控制和用药指导的理想分析方法。
[Objective] The reverse-phase high-performance liquid chromatography was used to analyze the fermentation liquid of pyrimidinemycin and the content of antifungal components was determined by external standard method. [Method] With DIKMA Diamonsil Ⅱ C18 reverse phase column and ultraviolet variable wavelength detector, the mobile phase was methanol - water (volume ratio is 20:80), the flow rate was 1 mL / min and the detection wavelength was 278 nm. [Result] The retention time of the antimycotic component of the amphotericin was 10.2 min, the linear correlation coefficient of the method was 0.999 8, the average recovery of the sample was 99.4% and the coefficient of variation was 0.25%. [Conclusion] This method is an ideal analytical method for the control of the fermentation and the guidance of the medication.
