目的探讨辛伐他汀和二苯基碘(DPI)对血管紧张素Ⅱ(AngⅡ)刺激的自发性高血压大鼠(SHR)外周血单个核细胞NADPH氧化酶2(NOX2)mRNA的表达和白细胞介素1β(IL-1β)分泌的影响。方法分离SHR外周血单个核细胞并进行培养,将细胞随机分为5组,每组6只:空白对照(仅加入培养液,A)组,AngⅡ(10-6mol/L,B)组,AngⅡ(10-6mol/L)+DPI(10-5mol/L,C)组,AngⅡ(10-6mol/L)+辛伐他汀(10-5mol/L,D)组,AngⅡ(10-6mol/L)+二甲基亚砜(DMSO,E)组。用RT-PCR检测各组单个核细胞NOX2 mRNA的表达,ELISA检测各组单个核细胞IL-1β蛋白的分泌。结果 NOX2 mRNA表达和IL-1β分泌,B组较A组明显升高(P<0.01),C、D组较B组均明显降低(P<0.01),E组与B组间差异无统计学意义。结论 AngⅡ促进SHR外周血单个核细胞IL-1β分泌与AngⅡ促进NOX2的表达有关;辛伐他汀抑制SHR外周血单个核细胞IL-1β的分泌可能与抑制其NOX2的表达有关。
Objective To investigate the effect of simvastatin and diphenyliodide (DPI) on the expression of NADPH oxidase 2 (NOX2) mRNA in peripheral blood mononuclear cells of spontaneously hypertensive rats (SHR) stimulated with angiotensin Ⅱ (Ang Ⅱ) The effect of hormone 1β (IL-1β) secretion. Methods Peripheral blood mononuclear cells (SHRs) were isolated and cultured. The cells were randomly divided into 5 groups with 6 mice in each group. Blank control group (group A), Ang Ⅱ (10-6mol / L, (10-6mol / L) + DPI (10-5mol / L, C), AngⅡ (10-6mol / L) + simvastatin ) + Dimethylsulfoxide (DMSO, E) group. The expression of NOX2 mRNA in each group of mononuclear cells was detected by RT-PCR. The secretion of IL-1β protein in mononuclear cells of each group was detected by ELISA. Results NOX2 mRNA and IL-1β secretion were significantly higher in group B than those in group A (P <0.01), while those in group C and D were significantly lower than those in group B (P <0.01). There was no significant difference between group E and group B significance. Conclusion AngⅡ can promote the secretion of IL-1β in peripheral blood mononuclear cells of SHR and the expression of NOX2 by AngⅡ. Simvastatin may inhibit the secretion of IL-1β in peripheral blood mononuclear cells of SHR, which may be related to the inhibition of NOX2 expression.
