目的观察转化生长因子β(TGF-β)抑制剂SB-505124对U87MG细胞放疗敏感性的影响。方法将U87MG细胞分为SB-505124处理组(A组)和对照组(B组),分别接受0、3、6、10和15Gy射线照射后,采用细胞计数试剂盒(CCK-8)检测细胞存活率,免疫荧光技术检测细胞损伤情况。结果 A组和B组射线照射后磷酸化H2AX荧光点数均较照射前增加[(9.02±1.96)个vs.(2.17±0.99)个和(8.75±2.07)个vs.(2.68±0.96)个](P<0.05);但A组与B组射线照射后细胞存活率、荧光点数均无统计学差异(P>0.05)。结论 SB-505124不能增加胶质母细胞瘤对放疗的敏感性。
Objective To observe the effect of transforming growth factor β (TGF-β) inhibitor SB-505124 on radiosensitivity of U87MG cells. Methods U87MG cells were divided into group A (SB-505124) and control group (group B). U87MG cells were irradiated with 0, 3, 6, 10 and 15Gy rays respectively and the cells were detected by CCK-8 Survival rate, immunofluorescence detection of cell damage. Results The number of phosphorylated H2AX fluorescence spots in group A and group B after irradiation increased by (9.02 ± 1.96) vs. (2.17 ± 0.99) and (8.75 ± 2.07) vs. (2.68 ± 0.96) before irradiation (P <0.05). However, there was no significant difference in cell viability and fluorescence spots between group A and group B after radiation (P> 0.05). Conclusion SB-505124 can not increase the sensitivity of glioblastoma to radiotherapy.
