目的:研究冠心宁片对急性心肌缺血犬冠脉流量的影响。方法:将48只Beagle犬随机分为伪手术组、模型对照组、冠心宁片高、中、低剂量组、阳性对照组。采用结扎冠状动脉左前降支的方法复制急性心肌缺血模型,观察犬的冠脉流量、心肌血流量、冠脉阻力,并检测结扎前及给药后30、60、120、180min时心肌缺血犬血浆一氧化氮(NO)含量。结果:冠心宁片各剂量组与模型组、伪手术组相比较,冠脉流量、心肌血流量增加(P<0.05),NO表达水平显著升高(P<0.05,P<0.01),冠脉阻力有效降低(P<0.05)。结论:冠心宁片主要通过增加冠脉流量、降低冠脉阻力、升高NO表达水平等环节发挥抗心肌缺血作用。
Objective: To study the effect of Guanxinning Tablet on coronary flow in dogs with acute myocardial ischemia. Methods: Forty-eight Beagle dogs were randomly divided into sham operation group, model control group, Guanxinning tablet high, middle and low dose group and positive control group. An acute myocardial ischemia model was created by ligation of the left anterior descending coronary artery. The coronary flow, myocardial blood flow, and coronary resistance were observed in dogs. Myocardial ischemia before and at 30, 60, 120, and 180 minutes after administration was measured. Dog plasma nitric oxide (NO) content. Results: Compared with the model group and the sham operation group, the coronary flow and myocardial blood flow of Guanxinning Tablets were increased (P<0.05), and the expression of NO was significantly increased (P<0.05, P<0.01). Pulse resistance was effectively reduced (P < 0.05). Conclusion: Guanxinning tablet can exert anti-ischemic effect mainly by increasing coronary flow, decreasing coronary resistance, and increasing NO expression level.
