目的:研究国产吸收性明胶海绵中明胶的交联程度,为进一步提高该产品质量和性能提供依据。方法:利用2,4,6-三硝基苯磺酸与肽链中赖氨酸侧链ε-氨基发生反应,生成在346nm波长处有最大吸收峰的三硝基苯衍生物,测定明胶与甲醛发生交联前后明胶蛋白质肽链赖氨酸侧链ε-氨基数量的变化,就可推知吸收性明胶海绵中明胶的交联程度。结果:利用此方法测得国产吸收性明胶海绵样品中明胶的交联度约为60%。结论:该方法可测定吸收性明胶海绵中明胶的交联度。
OBJECTIVE: To study the degree of cross-linking of gelatin in domestic absorbent gelatin sponge and provide the basis for further improving the quality and performance of the product. METHODS: 2,4,6-trinitrobenzene sulfonic acid was reacted with the ε-amino group of the lysine side chain in the peptide chain to produce the trinitrobenzene derivative with the maximum absorption peak at 346 nm. The gelatin and The degree of cross-linking of gelatin in absorbent gelatin sponge can be inferred by the change of ε-amino groups in the side chain of lysine of the gelatin protein peptide chain before and after the cross-linking of formaldehyde. Results: The degree of cross-linking of gelatin in domestic absorbent gelatin sponge samples measured by this method was about 60%. Conclusion: This method allows the determination of the degree of gelatin crosslinking in absorbent gelatin sponges.
