目的通过观察参仙升脉口服液及其有效成分对缓慢性心律失常大鼠cAMP及钙通道的影响,探讨其治疗缓慢性心律失常的作用机制。方法用普萘洛尔制备缓慢性心律失常大鼠的模型,观察参仙升脉口服液对缓慢性心律失常大鼠心肌细胞内cAMP浓度的影响。测定麻黄碱对大鼠心室肌细胞内钙离子浓度的影响。结果参仙升脉口服液及其有效成分能够提高缓慢性心律失常大鼠心肌细胞内cAMP的浓度,增强钙通道电流的密度。结论参仙升脉口服液通过cAMP-PKA-ICa信号通路来提升心率可能是其治疗缓慢性心律失常的作用之一。
Objective To observe the effects of Shenxian Shengmai Oral Liquid and its active ingredients on cAMP and calcium channels in rats with chronic arrhythmia and to explore its mechanism of action in the treatment of bradyarrhythmia. Methods The rat model of chronic arrhythmia was prepared by propranolol and the effect of Shenxian Shengmai Oral Liquid on cAMP concentration in rat cardiac myocytes was observed. Effect of ephedrine on intracellular calcium concentration in rat ventricular. Results Shenxian Shengmai Oral Liquid and its active ingredients can increase cAMP concentration in cardiomyocytes of rats with chronic arrhythmia and increase the density of calcium channel current. Conclusion Shenxian Shengmai oral solution through the cAMP-PKA-ICa signaling pathway to improve heart rate may be one of its role in the treatment of bradyarrhythmia.
