目的:研究苦杏仁中有效成分苦杏仁苷的最佳提取、精制方法。方法:采用正交实验法,以苦杏仁苷的含量为指标确定最佳提取条件;及不同结晶时间、乙醇用量对苦杏仁提取物精制的影响。结果:影响因素最大的是提取次数,其次是提取时间,再次是乙醇用量。最佳提取工艺确定为:6倍量95%乙醇提取2次,每次1h。所得粗品用无水乙醇(1∶30固液比)重结晶三次,得到含量为92.47%的苦杏仁苷纯品,苦杏仁苷转移率为45.92%。结论:此工艺步骤简单、经济可行,适用于苦杏仁苷的工业化生产。
Objective: To study the best extraction and purification of amygdalin, the active ingredient in bitter almonds. Methods: The optimum extraction conditions were determined by orthogonal test and the content of amygdalin. The effects of different crystallization time and ethanol dosage on the purification of extract of bitter almond were studied. Results: The most influential factors were the number of extraction, followed by the extraction time, and again the amount of ethanol. The best extraction process was determined as follows: 6 times the amount of 95% ethanol extraction 2 times, each time 1h. The crude product was recrystallized three times with absolute ethanol (1: 30 solid-to-liquid ratio) to obtain pure amygdalin content of 92.47%, and the amygdalin transfer rate was 45.92%. Conclusion: The process steps are simple, economical and feasible, suitable for the industrial production of amygdalin.
