目的:探讨复方金钱草颗粒对单侧输尿管梗阻(UUO)模型大鼠肾间质纤维化的影响。方法:将20只雄性SD大鼠随机分为对照组与治疗组,建立UUO模型,治疗组每天早、中、晚各给予复方金钱草颗粒1次,1周为1疗程;对照组则给予生理盐水。1周后处死大鼠,处死前留取大鼠的血液,取对侧肾脏行HE染色,观察肾脏组织的病理改变,并用免疫组化的方法检测大鼠肾脏组织中TGF-β1、III型胶原的表达,并检测大鼠血清中血肌酐、血尿酸的变化。结果:治疗组治疗后TGF-β1和III型胶原水平(0.39±0.06 ng/ml、15.3±4.8μg/L)均显著低于对照组(0.81±0.11 ng/ml、61.5±15.8μg/L)。治疗组治疗后尿素氮水平和肌酐水平(6.1±1.1 mmol/L、46.8±9.8μmol/L)均显著低于对照组(13.5±2.6 mmol/L、118.5±26.1μmol/L)。结论:复方金钱草在改善UUO大鼠中肾组织纤维化及改善肾功能有一定作用。
Objective: To investigate the effect of compound moneygloves on renal interstitial fibrosis in rats with unilateral ureteral obstruction (UUO). Methods: Twenty male Sprague-Dawley rats were randomly divided into control group and treatment group. UUO model was established. The treatment group was given 1, 2, brine. One week later, the rats were sacrificed and the blood of the rats was collected before sacrifice. The contralateral kidneys were stained with HE and the pathological changes of the kidneys were observed. The expressions of TGF-β1 and type III collagen in rat kidney were detected by immunohistochemistry The changes of serum creatinine and serum uric acid in rat serum were detected. Results: The levels of TGF-β1 and type III collagen in the treatment group (0.39 ± 0.06 ng / ml, 15.3 ± 4.8 μg / L) were significantly lower than those in the control group (0.81 ± 0.11 ng / ml, 61.5 ± 15.8 μg / L) . The levels of urea nitrogen and creatinine in the treatment group were significantly lower than those in the control group (6.1 ± 1.1 mmol / L, 46.8 ± 9.8 μmol / L) after treatment (13.5 ± 2.6 mmol / L, 118.5 ± 26.1 μmol / L). Conclusion: Pistacia chinense forsythia plays an important role in improving renal fibrosis and improving renal function in rats with UUO.
