针对罗汉果果实富含多糖和多酚类物质的特点,以广西罗汉果代表种质青皮果种的果实为材料,比较了改良Trizol法、改良异硫氰酸胍法、改良CTAB法和常规Trizol法4种不同的总RNA提取方法的提取效果。结果表明,改良Trizol法所提取的RNA呈现28S rRNA、18S rRNA和5S rRNA 3条清晰的谱带,且28S条带宽度接近18S的2倍,纯度高(D260/D280=2.01;D260/D230=2.02),完整性好(RIN=9.50),得率为(260.00±19.47)μg·g-1。RT-PCR结果进一步表明,改良Trizol法提取的RNA完全能够用于后续的分子生物学研究。
Aiming at the characters of polysaccharides and polyphenols in the fruit of Luohanguo fruit, the fruit of the representative species of Luohanguo in Guangxi was used as material. The improved Trizol method, modified guanidinium isothiocyanate method, modified CTAB method and conventional Trizol method were compared. Extraction of different kinds of total RNA extraction methods. The results showed that the RNA extracted by the modified Trizol method showed three distinct bands of 28S rRNA, 18S rRNA and 5S rRNA, and the 28S band width was nearly double that of 18S with high purity (D260 / D280 = 2.01; D260 / D230 = 2.02), good integrity (RIN = 9.50), the yield was (260.00 ± 19.47) μg · g-1. The results of RT-PCR further showed that the RNA extracted by the modified Trizol method can be completely used in subsequent molecular biology research.
