以人脑恶性胶质瘤细胞系 SHG-44为抗原免疫BALB/c 小鼠,通过杂交瘤技术获得了一株稳定地分泌抗胶质瘤细胞单克隆抗体的杂交瘤细胞株 SZ-39。经 ELISA、间接免疫荧光法和 ABC 免疫组化法分别检测了 SZ-39与各种肿瘤和正常细胞,脑瘤和正常人体组织的反应性。结果显示 SZ-39与10种被检测恶性胶质瘤细胞系中9种发生强结合反应,而与血液细胞等7种正常细胞及脑组织等12种人体组织无交叉反应。ABC 免疫组化检查了50例颅内肿瘤,结果提示 SZ-39与84.7%星形细胞瘤发生反应,尤以恶性星形细胞瘤反应最强。McAbSZ-39在移植性胶质瘤裸小鼠体内显示出良好的特异性。经免疫转移电泳法证实该抗体识别抗原为胶质瘤细胞膜上 Mw180000糖蛋白。
BALB / c mice were immunized with SHG-44 human glioblastoma cell line, and a hybridoma cell line SZ-39 which secreted monoclonal antibody against glioma cells was successfully obtained by hybridoma technique. The reactivity of SZ-39 with various tumor and normal cells, brain tumor and normal human tissues was detected by ELISA, indirect immunofluorescence and ABC immunohistochemistry respectively. The results showed that SZ-39 had a strong binding reaction with 9 of 10 kinds of malignant glioma cell lines, but no cross-reaction with 7 kinds of normal cells such as blood cells and 12 kinds of human tissues such as brain tissue. ABC immunohistochemical examination of 50 cases of intracranial tumors, the results suggest that SZ-39 and 84.7% astrocytoma reaction, especially strongest malignant astrocytoma. McAbSZ-39 showed good specificity in transplanted glioma nude mice. Immunofluorescence electrophoresis confirmed that the antibody recognition antigen is gl180000 glycoprotein on glioma cell membrane.
