以~3H-Dex为放射配基,采用受体单点结合分析法,检测了78例大肠癌患者的癌组织及68例正常肠段组织各组分糖皮质激素受体(GCR)的含量。~3H-Dex的浓度为12nmol/L,非放射配基Dex的浓度为放射配基~3H-Dex浓度的2000倍。结合和游离部分的分离采用滤膜法。滤膜烘干后用LKB液闪仪测量其放射性。蛋白浓度按Lowry法测定。78例大肠癌组织胞浆的GCR含量为3.25±4.89fmol/mg蛋白,33例癌胞膜为41.21
Using ~3H-Dex as radioligand, receptor single-site binding assay was used to detect the content of glucocorticoid receptor (GCR) in 78 cases of colorectal cancer and 68 cases of normal intestinal tissue. The concentration of ~3H-Dex is 12 nmol/L, and the concentration of non-radioactive ligand Dex is 2000 times that of radioactive ligand ~3H-Dex. The separation of the bound and free fractions was performed using a membrane filter method. The filter was dried and measured for radioactivity using a LKB liquid scintillator. The protein concentration was measured by the Lowry method. The cytoplasmic GCR content in 78 cases of colorectal cancer tissue was 3.25±4.89 fmol/mg protein, and the number of cancer cells in 33 cases was 41.21.
