目的研究海带多糖延缓皮肤衰老的作用机制。方法 SPF级昆明小鼠(8周龄)背部剃毛,随机分为对照组、模型组、海带多糖低、中、高剂量组和Vit E组。除对照组外,每日上、下午定时皮肤外涂相应药物。12个月后,取背部暴露皮肤,通过HE染色测量皮肤真皮层厚度,化学比色法检测皮肤羟脯氨酸(Hyp)含量,real time PCR法检测皮肤组织基质金属蛋白酶-1(MMP-1)mRNA、基质金属蛋白酶组织抑制物-1(TIMP-1)mRNA的表达水平,并检测皮肤超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)、过氧化氢(H2O2)的含量。结果与模型组比较,海带多糖可以显著增加真皮层厚度、皮肤Hyp含量、TIMP-1 mRNA表达水平及皮肤组织SOD、CAT、GSH-Px活性,显著降低皮肤MMP-1 mRNA表达水平及MDA、H2O2含量。结论海带多糖能够增加自然衰老小鼠皮肤胶原蛋白的含量,延缓皮肤衰老。其作用机制可能是通过减少氧自由基来调节皮肤基质金属蛋白酶的活性,从而影响皮肤胶原蛋白的含量有关。
Objective To study the mechanism of action of kelp polysaccharide in delaying skin aging. Methods SPF Kunming mice (8 weeks old) were shaved back and were randomly divided into control group, model group, low, medium and high dose of kelp polysaccharide group and Vit E group. In addition to the control group, the daily, afternoon skin outer coated with the appropriate drugs. Twelve months later, the exposed skin was taken from the back, and the thickness of the dermal layer was measured by HE staining. The content of hydroxyproline (Hyp) in skin was detected by chemical colorimetry. The expression of matrix metalloproteinase-1 ) mRNA and tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNA and protein expression were detected by immunohistochemistry. The expressions of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase GSH-Px) activity and malondialdehyde (MDA), hydrogen peroxide (H2O2) content. Results Compared with the model group, the kelp polysaccharide significantly increased the dermal layer thickness, the content of the skin Hyp, the expression level of TIMP-1 mRNA and the activity of SOD, CAT and GSH-Px in the skin tissue, and significantly reduced the expression of MMP-1 mRNA and MDA, H2O2 content. Conclusion Laminaria polysaccharide can increase the content of collagen in skin of mice with natural aging and delay skin aging. Its mechanism may be related to the reduction of oxygen free radicals to regulate the activity of the skin matrix metalloproteinase, thereby affecting the content of skin collagen.
