目的探讨烧伤患者体内磷酸化胰岛素样生长因子结合蛋白1(insulin-like growth factor binding protein-1,IGFBP-1)含量水平变化。方法选取2012年12月~2014年12月收治的86例烧伤患者为研究对象,收集烧伤患者自入院治疗至术后治疗10d的血浆,酶联免疫吸附法(ELISA)测定血浆中胰岛素、胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)和IGFBP-1含量,磷酸化IGFBP-1用Western Blotting检测。结果烧伤患者血浆IGFBP-1含量与总体表面积(TBSA)烧伤指数呈正相关,IGF-1含量随着TBSA值升高而显著下降(P<0.05)。在患者烧伤后72h内,IGF-1含量降低,而IGFBP-1含量[(184±26)ng/m L]升高,相比正常健康组IGFBP-1含量[(16±4)ng/m L]结果有显著统计学意义(P<0.05),同时血浆中可检测到磷酸化的IGFBP-1。结论烧伤患者血浆IGFBP-1含量变化和IGFBP-1持续的磷酸化与烧伤严重程度相关,IGFBP-1持续的磷酸化可抑制IGF的生物活性,并可作为判断烧伤患者预后的预警指标。
Objective To investigate the level of phosphorylated insulin-like growth factor binding protein-1 (IGFBP-1) in burn patients. Methods Eighty-six burn patients admitted from December 2012 to December 2014 were selected as the study subjects. Plasma samples were collected from hospitalized patients to 10 days post-treatment for burn patients. Serum insulin, insulin-like The levels of IGF-1 and IGFBP-1, phosphorylated IGFBP-1 were detected by Western Blotting. Results There was a positive correlation between the plasma IGFBP-1 level and the burn index of total body surface area (TBSA) in burn patients. The level of IGF-1 decreased significantly with the increase of TBSA value (P <0.05). The level of IGFBP-1 in IGFBP-1 was significantly higher than that in normal healthy group [(184 ± 26) ng / m L] within 72 hours after burn [16 ± 4 ng / m L] was statistically significant (P <0.05), meanwhile, phosphorylated IGFBP-1 was detected in plasma. Conclusion The changes of plasma IGFBP-1 level and the continuous phosphorylation of IGFBP-1 in burn patients are related to the severity of the burn. Persistent phosphorylation of IGFBP-1 may inhibit the biological activity of IGF and may be used as an early warning index to evaluate the prognosis of patients with burn.
