重组大肠杆菌ZJGSU01高效表达内切几丁质酶的培养条件

目的:研究重组大肠杆菌ZJGSU01高效表达内切几丁质酶的最优化培养条件。方法:以内切几丁质酶活力为指标,对诱导剂的诱导时间、诱导剂的添加量、装液量和接种量进行优化。结果:接种量5%,装液量100mL/500mL,重组菌培养1h,添加诱导剂IPTG的浓度为1.0mmol/L,有利于内切几丁质酶的表达。结论:通过上述条件的优化,可明显提高内切几丁质酶的表达量。 OBJECTIVE: To study the optimal culture conditions for efficient expression of endochitinase by recombinant E. coli ZJGSU01. Methods: The chitinase activity was used as an index to optimize the induction time, the amount of inducer, the volume of liquid and inoculum. Results: The amount of inoculum was 5%, the volume of liquid was 100mL / 500mL and the recombinant strain was cultured for 1h. The concentration of IPTG was 1.0mmol / L, which was favorable for the expression of endochitinase. Conclusion: The optimization of the above conditions can significantly increase the expression of endochitinase.
