目的:采用响应面法优化新疆黑果小檗根皮中总生物碱提取工艺。方法:以总生物碱提取率为考察指标,通过单因素试验,确定提取次数为3次,提取温度:80℃。采用响应面法优化乙醇浓度、提取时间、料液比3个因素。结果:得到最佳提取工艺为:乙醇体积分数50%,料液比1:51(g·m L~(-1)),提取时间2 h,提取次数3次,提取温度80℃。在该条件下,平行3次验证实验,总生物碱提取率为4.01%(RSD%=1.12%),可达到预测值的99.01%。结论:采用响应面法优化的黑果小檗根皮中总生物碱提取工艺稳定可行。
OBJECTIVE: To optimize the extraction process of total alkaloids in the root bark of barberry from the black berry of Xinjiang using response surface methodology. Methods: The extraction rate of total alkaloids was taken as the index of investigation. The number of extraction was determined by single factor test three times and the extraction temperature was 80 ℃. Response surface method was used to optimize the ethanol concentration, extraction time, the ratio of solid to liquid was 3 factors. Results: The optimal extraction process was as follows: the volume fraction of ethanol was 50%, the ratio of solid to liquid was 1:51 (g · m L -1), the extraction time was 2 h, the extraction times were 3 times and the extraction temperature was 80 ℃. Under this condition, the total alkaloid extraction rate was 4.01% (RSD% = 1.12%) in parallel to the three validation experiments, reaching 99.01% of the predicted value. Conclusion: The extraction process of total alkaloids in the root bark of Berberine bidentata optimized by response surface methodology is stable and feasible.
