目的:研究戊己丸药味配伍的特征图谱及其化学成分的变化规律。方法:采用毛细管电泳(HPCE)与反相高效液相色谱法对不同拆方进行考察,毛细管电泳以50 mmol·L-1的硼砂缓冲液-甲醇(2∶1)缓冲液为背景电解质,工作电压25k V,压力进样5 k Pa×15 s,电解质封口5 k Pa×10 s,柱温15℃。反相高效液相色谱法采用Agelapromosil C18色谱柱(4.6 mm×250 mm,5μm),柱温25℃,流速0.8 m L·min-1,流动相乙腈(A)-0.05 mol·L-1磷酸二氢钾(含0.1%磷酸)(B)进行梯度洗脱,检测波长230 nm,进样量5μL。采用撤药分析法对戊己丸进行研究,从定性定量两个角度进行评价,从整体上考察不同配伍情况方剂中化学成分的变化。结果:HPCE法确定了9个特征指纹峰,HPLC法确定27个特征指纹峰,其中黄连为该方君药,其生物碱类成分在不同配伍中变化较大;白芍中成分变化较小。结论:运用特征图谱技术结合定性、定量方法研究复方配伍,对复方新剂型的研制起到一定的指导作用。
OBJECTIVE: To study the characteristic map of compatibility of Wuji pills and its chemical composition. Methods: Different extraction methods were investigated by capillary electrophoresis (HPCE) and reversed-phase high performance liquid chromatography (RP-HPLC). Capillary electrophoresis was performed with 50 mmol·L -1 borax buffer-methanol Voltage 25k V, pressure injection 5kPa × 15s, electrolyte seal 5kPa × 10s, column temperature 15 ℃. The reversed-phase high performance liquid chromatography (RP-HPLC) was performed on an Agelapromosil C18 column (4.6 mm × 250 mm, 5 μm) at a flow rate of 0.8 m L · min- Potassium dihydrogen phosphate (containing 0.1% phosphoric acid) (B) gradient elution, detection wavelength of 230 nm, the injection volume 5μL. The drug withdrawal method was used to study the Wuji pills, and the qualitative and quantitative methods were used to evaluate the changes of the chemical components in different compatibility prescriptions. Results: There were 9 fingerprint peaks identified by HPCE method, and 27 fingerprint peaks were identified by HPLC method. The Coptis chinensis was the king herb and its alkaloids changed greatly in different compatibility. Conclusion: The combination of compound prescriptions with qualitative and quantitative methods using the technique of feature map plays a guiding role in the development of new dosage forms of compound prescriptions.
