本文以~(125)Ⅰ标记血管紧张素Ⅱ([~(125)Ⅰ]AⅡ)为配基,用放射分析法测定自发性高血压大鼠(SHR)脾淋巴细胞AⅡ受体,并与血压正常的WKA大鼠进行对比观察。结果表明两种大鼠淋巴细胞在pH 7.6,22℃条件下与[~(125)Ⅰ]AⅡ发生特异性结合。并可被非标记AⅡ及AⅡ拮抗剂(Sar~1,Ile~8]AⅡ竞争抑制。经Scatchard plot分析,提示两种大鼠脾淋巴细胞上均可能存在AⅡ的高、低两种亲和力受体,且SHR淋巴细胞AⅡ受体的亲和力高于WKA大鼠。体外实验还证明AⅡ抑制大鼠淋巴细胞增殖及IL-2的产生。提示SHR的免疫功能抑制可能与淋巴细胞表达高亲和力的AⅡ受体有关。
In this study, angiotensin Ⅱ ([~ (125) Ⅰ] AⅡ) was used as a ligand for 125 Ⅰ in the study. The AⅡ receptor of splenic lymphocytes in spontaneously hypertensive rats (SHR) Normal WKA rats were compared. And can be competitively inhibited by non-labeled AⅡ and AⅡ antagonists (Sar ~ 1, Ile ~ 8] AⅡ.According to Scatchard plot analysis, both high and low affinity AII receptors of AⅡ may exist on splenic lymphocytes , And the affinity of A receptor of SHR lymphocytes was higher than that of WKA rats.In vitro experiments also showed that AⅡ inhibited the proliferation of rat lymphocytes and the production of IL-2, suggesting that the immune function inhibition of SHR may be related to the high affinity AⅡ receptor Body related.
