目的利用Meta分析的方法,综合评价血管紧张素转换酶(ACE)基因插入/缺失(I/D)多态性与乳腺癌易感性的相关性。方法计算机检索Pubmed、Embase、中国生物医学文献数据库和万方数据库等,并手工检索相关杂志,收集有关不同人群ACE基因I/D位点单核苷酸多态性的等位基因频率、基因型频率与乳腺癌易感性相关性的研究。检索时间截止至2014年8月。在评价纳入研究并提取有效数据后,采用Stata 12.0软件进行Meta分析。选择的遗传模型包括:DI+II与DD比较,II与DI+DD比较,DD与II比较以及等位基因D与I之间比较。结果共10篇文献(13项病例对照研究)符合既定的纳入标准,合计10 405例研究对象。Meta分析结果显示:在全部人群中,ACE基因I/D位点的等位基因频率、基因型分布与乳腺癌易感性无相关性。根据种族不同进行亚组分析结果显示:I/D位点等位基因频率及基因型分布与乳腺癌易感性的相关性在黄种人、美洲人和高加索人群中差异均无统计学意义(P>0.05)。结论 Meta分析结果提示,ACE基因I/D多态性与乳腺癌易感性可能不具有相关性。但目前的结论尚需进一步大样本、高质量的研究去验证。
Objective To evaluate the relationship between angiotensin converting enzyme (ACE) gene insertion / deletion (I / D) polymorphism and susceptibility to breast cancer using Meta-analysis. Methods Pubmed, Embase, China Biomedical Literature Database and Wanfang Database were searched by computer. The related journals were searched by hand, and the frequencies of alleles and genotypes of ACE gene I / D SNPs in different populations were collected. Study on the correlation between frequency and susceptibility to breast cancer. The search time is up to August 2014. After evaluating the inclusion studies and extracting valid data, Meta analysis was performed using Stata 12.0 software. Selected genetic models include: DI + II vs. DD, II vs. DI + DD, DD vs II, and alleles D vs. I. Results A total of 10 articles (13 case-control studies) met the established inclusion criteria and totaled 10,405 subjects. Meta-analysis showed that allele frequency and genotype distribution of ACE gene I / D locus had no correlation with susceptibility to breast cancer in all populations. Subgroup analysis based on different races showed that there was no significant difference in the frequencies of alleles and genotype distribution between I / D loci and the susceptibility to breast cancer among Yellow, American and Caucasian populations (P> 0.05). Conclusions The results of Meta analysis suggest that ACE gene I / D polymorphism may not be correlated with breast cancer susceptibility. However, the current conclusion still needs further large sample and high quality research to verify.
