目的:观察天王补心丸四个SP825大孔吸附树脂洗脱部分的镇静催眠作用。方法:实验于2005-06/08在军事医学科学院放射与辐射医学研究所药理毒理研究室完成。健康昆明种小鼠200只,随机数字表法分为空白对照组,阳性药组,水洗脱部位组,体积分数为0.2乙醇洗脱部位组,体积分数为0.5乙醇洗脱部位组,体积分数为0.8乙醇洗脱部位组,每组10只。空白对照组给予等体积生理盐水,阳性药组给予阳性药,其余四个受试药物组分别给予天王补心丸SP825洗脱物。①协同睡眠试验:各组末次给药后30min,腹腔注射戊巴比妥钠40mg/kg,以小鼠翻正反射消失达1min以上为入睡指标,记录睡眠时间。②自发活动试验:分别于各组末次给药30min、90min两个时间点观测小鼠自发活动,记录5min的自发活动次数。③抗惊厥试验:各组末次给药后1h,腹腔注射戊四唑溶液(65mg/kg),以小鼠前肢和头部发生阵挛性抽搐为指标,记录惊厥潜伏期。结果:实验大鼠200只均进入结果分析。①协同睡眠试验结果:天王补心丸SP825体积分数为0.2乙醇洗脱部分能够明显延长阈剂量戊巴比妥钠引起小鼠睡眠的时间(P<0.05)。②自发活动试验结果:给药后30min,各受试药物组的自发活动与对照组相比均无显著性差异(P>0.05);给药后90min,天王补心丸SP825体积分数为0.5乙醇洗脱部分能够明显减少小鼠的自发活动次数(P<0.05)。③抗惊厥试验结果:天王补心丸SP825体积分数为0.2和0.5乙醇洗脱部分均能够明显延长戊四唑所致小鼠阵挛性抽搐的潜伏期(P<0.05)。结论:天王补心丸镇静催眠的活性部位是其亲水性较强的SP825体积分数为0.2和0.5乙醇洗脱部分。
Objective: To observe the sedative and hypnotic effects of the fractions eluted by four SP825 macroporous adsorption resins from Tianwang Buxin Pills. METHODS: The experiment was performed at the Department of Pharmacology and Toxicology, Institute of Radiation and Radiation Medicine, Academy of Military Medical Sciences from June 2005 to August 08. 200 healthy Kunming mice were randomly divided into blank control group, positive drug group, water elution site group, volume fraction of 0.2 ethanol elution site group, volume fraction of 0.5 ethanol elution site group, volume fraction A group of 0.8 ethanol-eluting sites, 10 in each group. The control group was given an equal volume of normal saline, the positive drug group was given a positive drug, and the remaining four drug groups were given Tianwang Buxin Pill SP825 eluate. 1 Synergy sleep test: 30 minutes after the last administration of each group, pentobarbital sodium 40 mg/kg was injected intraperitoneally, and the righting reflex disappeared for more than 1 min as the index of sleep, and the sleep time was recorded. 2 Spontaneous activity test: Spontaneous activities of the mice were observed at the time points of 30 min and 90 min after the last administration in each group, and the number of spontaneous activities was recorded for 5 min. 3 anticonvulsant test: 1h after the last administration of each group, intraperitoneal injection of pentylenetetrazol solution (65mg/kg), the mouse forelimb and head clonic seizures as indicators, recorded convulsive latency. RESULTS: A total of 200 experimental rats were involved in the result analysis. 1 Synergistic sleep test results: Tianwangbuxin pill SP825 volume fraction 0.2 ethanol elution part can significantly prolong the time when the threshold dose of pentobarbital sodium causes mice to sleep (P<0.05). 2 Spontaneous activity test results: 30 min after administration, spontaneous activity of each test group was not significantly different from that of the control group (P>0.05); 90 min after administration, the volume of Tianwang Buxin Pill was 0.5 0.5 ethanol. The eluted fraction significantly reduced the number of spontaneous movements of mice (P<0.05). 3 Anticonvulsant test results: Tianwangbuxin pill SP825 volume fraction of 0.2 and 0.5 ethanol elution part can significantly prolong the incubation period of pentylenetetrazole induced clot convulsions in mice (P<0.05). Conclusion: The active site of sedative and hypnotic effect of Tianwang Buxin Pill is its strong hydrophilic SP825 volume fraction of 0.2 and 0.5 ethanol fractions.
