目的:探讨灯盏花素对糖尿病大鼠心肌的干预作用及对神经肽Y(NPY)水平的影响。方法:建立T1DM大鼠模型,将成模后的成年Wister大鼠随机分为糖尿病模型组、胰岛素治疗组、灯盏花素低剂量干预组、灯盏花素高剂量干预组,另设正常对照组。观察血糖、体重、心脏重量指数、心系数,放免法测定NPY水平,常规HE染色观察心肌组织的形态学改变。结果:糖尿病大鼠心脏重量指数、心系数以及血浆、心肌组织内NPY水平显著升高,HE染色显示心肌细胞存在等多种病理改变。胰岛素治疗与灯盏花素干预后均能改善糖尿病大鼠心肌组织的病理变化,其中灯盏花素高剂量干预效果同胰岛素治疗组。在干预和治疗过程中血浆NPY水平降低,心肌组织NPY水平升高。结论:糖尿病大鼠血浆NPY水平增高,灯盏花素可以降低血浆NPY水平,改善糖尿病大鼠心功能、心肌细胞肥大现象,并可改善糖尿病大鼠的一般生理状况。
Objective: To investigate the effects of breviscapine on myocardium in diabetic rats and its effect on neuropeptide Y (NPY) levels. Methods: The adult Wistar rats were randomly divided into diabetic model group, insulin treatment group, breviscapine low-dose intervention group, breviscapine high-dose intervention group, and another normal control group. The blood glucose, body weight, heart weight index, cardiac index, radioimmunoassay were used to measure the level of NPY. The morphology of myocardial tissue was observed by routine HE staining. Results: The cardiac weight index, cardiac index and the level of NPY in plasma and myocardium of diabetic rats were significantly increased. HE staining showed the presence of cardiomyocytes and other pathological changes. Insulin treatment and breviscapine intervention can improve the pathological changes of myocardial tissue in diabetic rats, of which Breviscapine high-dose intervention effect with the insulin treatment group. During the intervention and treatment of plasma NPY levels decreased myocardial NPY levels. Conclusion: The plasma NPY level in diabetic rats increased. Breviscapine can reduce plasma NPY levels, improve cardiac function and myocardial hypertrophy in diabetic rats, and improve the general physiological status of diabetic rats.
