本实验用Wistar雄性大鼠肺泡巨噬细胞(PAM)与Sb_2O_2共同体外培养。透射电镜下可见PAM变圆,伪足消失,溶酶体数量早期增多、后期减少并呈大颗粒状,线粒体肿胀,常染色质减少,异染色质增多,核固缩,最后细胞崩解。培养基中酸性磷酸酶(ACP)和乳酸脱氢酶(LDH)含量早期明显增高,分别与空白组有显著差异(均P<0.01),并呈持续上升趋势。提示Sb_2O_2对PAM有较肯定的细胞毒作用,可速成膜性结构明显损伤。
In this experiment, Wistar male rat alveolar macrophages (PAM) and Sb 2 O 2 co-culture in vitro. Under the transmission electron microscope, PAM became round and pseudopodia disappeared. The number of lysosomes increased at early stage, decreased later and presented as large granules, swelling of mitochondria, decrease of euchromatin, increase of heterochromatin, nuclear pyknosis and finally disintegration of cells. The content of acid phosphatase (ACP) and lactate dehydrogenase (LDH) in the culture medium were significantly increased at early stage, which were significantly different from the blank group (all P <0.01), and continued to increase. It indicated that Sb 2 O 2 had a positive cytotoxic effect on PAM, which could cause obvious damage to the membranous structure.
