建立辣木油中油酸的UPLC-MS/MS测定方法.采用Waters液质联用仪,色谱柱为Acquity UPLC BEH C18柱(50 mm×2.1 mm,1.7μm);以0.1%甲酸溶液-乙腈(5∶95)为流动相,柱温40℃;流速0.4 m L/min;ESI+,多反应监测模式(MRM),监测离子为m/z 284.321 9→m/z46.025 7(定量),m/z 284.321 9→m/z 43.034 3和m/z 284.321 9→m/z 57.078 9(定性).标准曲线为Y=2.027 23×108X-128 016(r=0.999 642),油酸在0.262 48~6.562μg/m L内线性关系良好,平均回收率为97.45%.本方法简便、快速,结果准确,可用于辣木油的质量控制.
A UPLC-MS / MS method was developed for the determination of oleic acid in Moringa oleracea L. Using a Waters LC / MS instrument, the column was Acquity UPLC BEH C18 (50 mm × 2.1 mm, 1.7 μm); the column was eluted with 0.1% formic acid in acetonitrile 5:95) as the mobile phase, the column temperature was 40 ℃, the flow rate was 0.4 m L / min, the ESI + and multi-reaction monitoring mode was MRM, the monitored ions were m / z 284.321 9 → m / z 46.025 7 / z 284.321 9 → m / z 43.034 3 and m / z 284.321 9 → m / z 57.078 9. The standard curve is Y = 2.027 23 × 108X-128 016 (r = 0.999 642) and oleic acid is 0.262 48 ~ 6.562μg / m L, the average recovery was 97.45% .The method is simple, rapid, accurate and can be used for the quality control of Moringa oleifera oil.
