目的:探讨miR-19a在去势抵抗性前列腺腺癌中的表达及其对去势抵抗性前列腺癌细胞株PC3及DU145的增殖及凋亡的影响。方法:用定量PCR方法检测10例去势抵抗性前列腺癌细胞组织及相关癌旁的miR-19a表达,通过MTT、克隆形成、Western blotting、流式细胞仪检测增殖及凋亡及体内成瘤的方法来研究miR-19a对于去势抵抗性前列腺癌细胞增殖及凋亡的影响。结果:miR-19a在去势抵抗性前列腺癌组织中的表达相对于癌旁组织显著提高。抑制miR-19a在激素非依赖前列腺癌中的表达,可抑制细胞的增殖及集落形成并增加其凋亡。结论:在激素非依赖性前列腺癌中,miR-19a起着癌基因作用,与前列腺癌的增殖及凋亡相关。
Objective: To investigate the expression of miR-19a in ovariectomized prostate cancer and its effect on the proliferation and apoptosis of ovarian cancer cell lines PC3 and DU145. Methods: The expression of miR-19a was detected by quantitative PCR in 10 cases of ovariectomized prostate cancer tissues and related cancerous tissues. MTT, colony formation, Western blotting and flow cytometry were used to detect the proliferation and apoptosis of tumor cells in vivo and in vivo Methods To study the effects of miR-19a on the proliferation and apoptosis of ovarian cancer cells in ovariectomized prostate cancer. Results: The expression of miR-19a in ovariectomized prostate cancer tissues was significantly higher than that in paracancerous tissues. Inhibition of miR-19a expression in hormone-independent prostate cancer can inhibit cell proliferation and colony formation and increase its apoptosis. Conclusion: miR-19a plays an oncogene role in hormone-independent prostate cancer and is associated with prostate cancer proliferation and apoptosis.
