目的:通过观察TGF-β1在哮喘大鼠模型中动态变化,并用柴朴汤进行干预,探讨柴朴汤在哮喘大鼠中的治疗作用,为哮喘的预防及治疗提供进一步的思路。方法:以卵清白蛋白致敏及反复激发复制哮喘大鼠模型,常规切片观察气道内管壁厚度及气道平滑肌厚度;用RT-PCR检测TGF-β1mRNA的表达;用免疫组化法检测TGF-β1蛋白的表达。结果:大鼠哮喘激发后2周出现气道平滑肌增厚,并且随着激发时间的延长,气道平滑肌逐渐增厚。同时,哮喘大鼠支气管肺组织中TGF-β1mRNA及TGF-β1蛋白表达随着激发时间的延长而逐渐升高,呈现出一定的动态变化。用柴朴汤干预后,TGF-β1的表达下调。结论:在哮喘气道重塑过程中,TGF-β1的表达随着激发时间的延长逐渐增加,是一个动态变化的过程,柴朴汤抑制气道重塑的机制可能与下调TGF-β1的表达有关。
OBJECTIVE: To observe the dynamic change of TGF-β1 in asthmatic rat model and to explore the therapeutic effect of Chai Pu-Tang in asthmatic rat by using Chai Pu-Tang intervention and to provide further ideas for the prevention and treatment of asthma. Methods: Asthmatic rat model was induced by ovalbumin sensitization and repeated stimulation. The thickness of airway wall and the thickness of airway smooth muscle were observed by routine sections. The expression of TGF-β1 mRNA was detected by RT-PCR. The expression of TGF- β1 protein expression. Results: Airway smooth muscle thickening occurred 2 weeks after asthma in rats, and the airway smooth muscle gradually thicken with the prolongation of time. Meanwhile, the expression of TGF-β1mRNA and TGF-β1 protein in bronchial lung tissue of asthmatic rats increased gradually with the prolongation of excitation time, showing a certain dynamic change. With Chai Pu Tang intervention, the expression of TGF-β1 down. Conclusion: The expression of TGF-β1 gradually increases with the prolongation of the time of asthma airway remodeling, which is a dynamic process. Chai Pu Tang inhibits airway remodeling and may down-regulate the expression of TGF-β1 related.
