目的探讨大鼠脑挫伤后HIF-1a蛋白表达的规律。方法参照Feeney,s法建立大鼠脑挫伤模型,分别于伤后2h,6h,12h,24h,2d,3d,5d,7d,14d取脑,运用免疫组化SABC法观察HIF-1a蛋白的表达。结果对照组偶见阳性反应细胞。损伤后2h,即可观察到HIF-1a蛋白表达,6h后表达明显增强,24h达高峰,随后阳性反应细胞逐渐减少,7d后仍可见少量表达,14d后恢复。表达呈弥漫性,以大脑皮层、海马、脑干最为明显,不同部位的变化趋势一致。各实验组与对照组比较有统计学意义(P<0.05)。结论HIF-1a蛋白的表达规律可用于脑挫伤的鉴定及损伤经过时间的推断。
Objective To investigate the regularity of HIF-1a protein expression in rats after brain contusion. Methods The brain contusion model was established by Feeney’s method. The brain was taken at 2h, 6h, 12h, 24h, 2d, 3d, 5d, 7d and 14d after injury respectively. The expression of HIF-1a protein was detected by immunohistochemical SABC method . Results The control group occasionally seen positive cells. The expression of HIF-1a protein was observed 2 h after injury, and the expression of HIF-1a protein was observed 6 h after the injury. The expression of HIF-1a protein reached its peak at 6 h and then gradually decreased. The expression was diffuse to the cerebral cortex, hippocampus, brainstem most obvious changes in different parts of the same trend. The experimental group and the control group were statistically significant (P <0.05). Conclusion The expression pattern of HIF-1a protein can be used to identify cerebral contusion and infer the time course of injury.
