目的:建立SHR大鼠脑组织中天麻苷元的含量测定方法,考察天麻与钩藤配伍前后效应成分天麻苷元在脑组织中的分布情况。方法:采用HPLC测定脑组织中天麻苷元含量,流动相乙腈-0.03%磷酸溶液(7∶93),检测波长221 nm。检测天麻与钩藤配伍前后效应成分天麻苷元在脑组织中的分布情况。结果:大鼠血浆中天麻苷元的线性范围0.5~100 mg·L-1(R2=0.999 9),方法回收率>74.89%,准确度>98%。配伍前、后天麻苷元在大鼠脑组织的药代动力学参数Cmax分别为0.485,0.373μg·g-1,AUC依次为111.366,213.585μg·g-1·min。结论:该方法能准确、灵敏测定大鼠脑组织中天麻苷元含量。天麻与钩藤配伍对天麻苷元在SHR大鼠脑组织中分布具有先抑制后促进影响。
OBJECTIVE: To establish a method for determining the content of gastrodin in brain tissue of SHR rats, and to study the distribution of the components of gastrodin in brain tissue before and after the combination of Gastrodia elata Blume and Uncaria. Methods: The content of gastrodin in brain tissue was determined by HPLC. The mobile phase was acetonitrile - 0.03% phosphoric acid solution (7:93). The detection wavelength was 221 nm. Detection of Gastrodia elata and Uncaria compatibility effect component before and after distribution of the content of spine aglycone in brain tissue. Results: The linear range of spiramycin in rat plasma was 0.5-100 mg · L-1 (R2 = 0.999 9). The recovery rate was> 74.89% and the accuracy was> 98%. Before and after compatibility, the pharmacokinetic parameters Cmax of post-apis melliferae in rat brain were 0.485,0.373μg · g-1, and the AUCs were 111.366,213.585μg · g-1 · min. Conclusion: This method can accurately and sensitively determine the content of gastrodin in rat brain tissue. Gastrodia elata and Uncaria rhynchophylla compatibility has a suppressive effect on the distribution of gastrodin in brain tissue of SHR rats.
