以花楸树离体成熟和未成熟合子胚为外植体,研究了不同产地、不同发育时间及不同植物生长调节剂组合对花楸树愈伤组织诱导和体胚发生的影响。结果表明:以成熟合子胚为外植体,MS+2.0mg/L NAA+1.5mg/L 6-BA对诱导愈伤组织和体胚发生效果好;不同产地合子胚的体胚诱导率排序为敦化>哈尔滨>五营>山河。以未成熟合子胚为外植体,MS+0.05mg/L 2,4-D+1.0mg/L 6-BA愈伤诱导率高,体胚发生在MS+0.2mg/L 2,4-D+1.0mg/L6-BA最适宜;不同产地合子胚的体胚诱导率排序为延吉>哈尔滨>伊春。
In vitro maturation and immature zygotic embryos of Catalpa bungei were used as explants to study the effects of different producing areas, different developmental stages and combinations of different plant growth regulators on callus induction and somatic embryogenesis. The results showed that the induction of callus and somatic embryo by MS + 2.0mg / L NAA + 1.5mg / L 6-BA was better than mature zygotic embryos. The induction rate of somatic embryos from different regions was Dunhua> Harbin> Five Camps> Mountain River. Immature zygotic embryos as explants, MS + 0.05mg / L 2,4-D + 1.0mg / L 6-BA callus induction rate, somatic embryogenesis in MS + 0.2mg / L 2,4-D + 1.0mg / L 6-BA was the most suitable. The order of somatic embryo induction of zygotic embryos from different origins was Yanji> Harbin> Yichun.
