目的:探讨红景天、蛹虫草、大黄配伍(中药复方FF16)改善IRF小鼠葡萄糖代谢紊乱的作用机制。高脂饮食诱导C57BL/6J小鼠形成胰岛素抵抗合并糖耐量低减的IRF小鼠,分为模型对照组IRF、罗格列酮(Rosi)组和FF16组。以禁食血糖水平测定、腹腔注射葡萄糖耐量试验(IPGTT)评价动物的糖代谢状态;采用胰岛素耐量试验(ITT)、血胰岛素测定和稳态模型胰岛素抵抗指数(HOMA-IR)评价机体的胰岛素敏感性。应用Western blot方法,检测小鼠肝脏胰岛素信号通路相关蛋白Akt,p-Akt,糖代谢相关蛋白GSK3β,p-GSK3β的表达。结果显示,中药复方FF16能够改善IRF小鼠的糖代谢紊乱,使葡萄糖耐量试验AUCIPGTT较IRF组降低22.3%;能显著增加IRF小鼠的胰岛素敏感性,使胰岛素耐量试验AUCITT降低22.1%,HOMA-IR指数降低49.5%。FF16使IRF小鼠肝脏Akt磷酸化水平升高116.4%,GSK3β磷酸化水平升高24.9%。推测中药复方FF16可通过增强IRF小鼠肝脏糖原合成,改善机体糖代谢紊乱状态。
Objective: To investigate the mechanism of Rhodiola, Cordyceps militaris, rhubarb compatibility (FF16) to improve glucose metabolism in IRF mice. High-fat diet induced IRF mice with insulin resistance and impaired glucose tolerance in C57BL / 6J mice, which were divided into model control group (IRF), rosiglitazone (Rosi) group and FF16 group. The fasting glucose level was measured and the glucose metabolism test was performed by intraperitoneal injection of glucose tolerance test (IPGTT). The insulin sensitivity was evaluated by insulin tolerance test (ITT), insulin and insulin homeostasis model (HOMA-IR) Sex. The expression of Akt, p-Akt and GSK3β, p-GSK3β in hepatic insulin of liver were detected by Western blot. The results showed that FF16 could improve the disorder of glucose metabolism in IRF mice and decrease the AUCIPGTT of glucose tolerance test by 22.3% compared with IRF group. It could significantly increase the insulin sensitivity of IRF mice and decrease the AUCITT of insulin resistance test by 22.1% The IR index dropped 49.5%. FF16 increased the level of Akt phosphorylation and the GSK3β phosphorylation in IRF mice by 24.9% and 116.4%, respectively. It is speculated that FF16 can improve the glycometabolism of liver in IRF mice by improving glycogen synthesis.
