目的研究慢性前列腺炎SD大鼠前列腺组织Smad4和Smad7的表达,并初步探讨它对慢性前列腺炎发病机制的意义。方法将40只6月龄SD大鼠随机分为正常对照组和CP实验组,每组20只。实验组采用去势+高剂量苯甲酸雌二醇肌注方法建模,并以免疫组化和蛋白免疫印迹实验方法检测Smad4、Smad7在前列腺组织中的表达变化。结果与正常对照组相比,Smad7表达显著下调(P<0.01),Smad4表达亦显著下调(P<0.01);Smad4与Smad7在慢性前列腺炎大鼠前列腺组织中表达无相关性。结论Smad4、Smad7在慢性前列腺炎前列腺组织中表达异常,提示可能参与CP的病理过程。
Objective To investigate the expression of Smad4 and Smad7 in prostate tissue of SD rats with chronic prostatitis and to explore its significance on the pathogenesis of chronic prostatitis. Methods Forty six-month-old SD rats were randomly divided into normal control group and CP experimental group, with 20 rats in each group. The experimental group was treated by castration + high-dose estradiol benzoate intramuscular injection. The expression of Smad4 and Smad7 in prostate tissue was detected by immunohistochemistry and Western blotting. Results Compared with the normal control group, Smad7 expression was significantly down-regulated (P <0.01) and Smad4 expression was also significantly down-regulated (P <0.01). There was no correlation between the expression of Smad4 and Smad7 in prostatic tissues of chronic prostatitis rats. Conclusion Smad4 and Smad7 are abnormally expressed in prostatic tissue of chronic prostatitis, suggesting that they may be involved in the pathological process of CP.
