目的构建并鉴定β1-肾上腺素受体细胞外第二环的表达载体。方法应用聚合酶链技术(PCR),从大鼠基因组中扩增出目的基因,与表达载体pGEX-4T-1重组,采用PCR、限制性内切酶酶切及DNA测序进行鉴定。结果本实验通过采用基因克隆技术构建了β1-肾上腺素受体细胞外第二环的表达载体,并对其进行了鉴定。结论本实验为以后表达该功能肽段和研究相应抗体的致病机制奠定了基础。
Objective To construct and identify the extracellular extracellular β1-adrenergic receptor expression vector. Methods Polymerase chain reaction (PCR) was used to amplify the target gene from the rat genome and recombined with the expression vector pGEX-4T-1. PCR, restriction endonuclease digestion and DNA sequencing were used to identify the target gene. Results In this experiment, the expression vector of β1-adrenoceptor extracellular loop 2 was constructed by gene cloning technique and identified. Conclusion This experiment laid the foundation for the future expression of the functional peptide and the study of the pathogenesis of the corresponding antibodies.
