用荧光偏振法测定荧光素的荧光寿命的灵敏度和准确度较高。在荧光素钠荧光强度最强时——激发波长490nm,发射波长515nm、pH值8.5、溶液浓度1.6×10~(-5)mol/L,测定荧光素钠溶液的荧光偏振度,测定不同温度时荧光素钠溶液的粘度,再根据Perrin公式,求出荧光素钠的荧光寿命。用荧光偏振法测得荧光素钠荧光寿命为3.4ns、p<0.02,与文献值符合良好,结果令人满意。
The sensitivity and accuracy of the fluorescence lifetime determination of fluorescein by fluorescence polarization are high. When fluorescence intensity of sodium fluorescein was the strongest-the excitation wavelength was 490nm, the emission wavelength was 515nm, the pH value was 8.5 and the concentration of solution was 1.6 × 10 -5 mol / L, the fluorescence polarization degree of sodium fluorescein solution was measured. When the viscosity of sodium fluorescein solution, and then according to Perrin formula, find out the fluorescence lifetime of fluorescein sodium. Fluorescence polarization measured fluorescence lifetime of sodium fluorescein 3.4ns, p <0.02, with the literature values in good agreement, the results are satisfactory.
