目的通过体外实验,观察不同体积分数一氟二氯乙烷(F-141b)对中国仓鼠肺成纤维细胞(V79细胞)的毒性作用。方法用不同体积分数(10、20、30、40、50和60 ml/L)且经抽滤的一氟二氯乙烷对V79细胞进行染毒,分别培养不同时间(3、6、12、24和48 h)后,于显微镜下计数存活细胞数,计算存活率和IC50。同时设阴性对照组。结果在任一培养时间段,细胞存活率均随染毒剂量增加而降低,短时间组(3 h、6 h)和长时间组(24 h、48 h)组间比较差别具统计学意义(P<0.05);染毒剂量相同,细胞存活率随时间增加而降低,各染毒剂量组间差别显著(P<0.05),其中3 h组、12 h组和48 h组相关具有显著的统计学意义(P<0.05)。存在明显的剂量―效应和时间―效应关系。24 h染毒IC50体积分数为32.37 ml/l。结论在本实验条件下,一氟二氯乙烷具有显著的细胞毒性作用,并存在剂量和时间效应。
Objective To observe the toxic effects of different concentrations of F-141b on Chinese hamster lung fibroblasts (V79 cells) in vitro. Methods V79 cells were exposed to different concentrations of fluvastatin (10, 20, 30, 40, 50 and 60 ml / L) and filtered with fluorodichloroethane and incubated at different times (3, 24 and 48 h), the number of viable cells was counted under a microscope to calculate the survival rate and the IC50.
