目的观察淫羊藿总黄酮注射液(EFI)对H2O2诱导心肌细胞损伤的保护作用。方法通过体外培养大鼠乳鼠心肌细胞,MTT法检测心肌细胞增殖状况,流式细胞仪检测心肌细胞凋亡,透射电镜观察心肌细胞超微结构。结果H2O2可剂量依赖性增加对心肌细胞增殖的抑制率,100μmol/LH2O2作用12h后心肌细胞凋亡率为(23.94±3.52)%,高于正常对照组的(1.98±1·22)%;心肌超微结构呈凋亡特征性变化;100、200、400mg/LEFI可剂量依赖性降低心肌细胞抑制率,显著降低心肌细胞凋亡率,分别为(15.12±3.61)%、(9.38±3.14)%和(4.49±0.52)%;且能改善心肌细胞超微结构变化。结论淫羊藿总黄酮注射液对H2O2诱导的心肌细胞损伤具有保护作用。
Objective To observe the protective effect of epimedium total flavonoids injection (EFI) on H2O2-induced cardiomyocyte injury. Methods Rat cardiomyocytes were cultured in vitro. The proliferation of cardiomyocytes was detected by MTT assay. Cardiomyocyte apoptosis was detected by flow cytometry. The ultrastructure of cardiomyocytes was observed by transmission electron microscope. Results H2O2 dose-dependently increased the inhibition rate of cardiomyocyte proliferation. The apoptosis rate of cardiomyocytes was (23.94±3.52)% after 100 μmol/LH2O2 for 12 h, which was higher than that of the control group (1.98±1·22)%; The ultrastructure showed apoptotic characteristic changes; 100, 200, 400mg/LEFI dose-dependently reduced myocardial cell inhibition rate, and significantly reduced the rate of cardiomyocyte apoptosis, (15.12±3.61)%, (9.38±3.14)%, respectively. And (4.49 ± 0.52)%; and can improve the ultrastructural changes of myocardial cells. Conclusion Epimedium flavonoids injection can protect H2O2-induced cardiomyocyte injury.
