为了比较不同转染方式的转染效率,本文利用针对HBVS基因和e基因的反义RNA真核表达载体,分别以磷酸钙法和脂质体法转染人肝癌细胞系HepG2.2.15,通过RIA方法检测转染3天后细胞培养液中HBsAg和HBeAg的表达水平,以观察内源性反义RNA对HBV基因表达的抑制作用。研究证实脂质体法转染效率更高,说明脂质体是较理想的基因治疗运载方式。
In order to compare the transfection efficiency of different transfection methods, antisense RNA eukaryotic expression vector targeting HBVS gene and e gene was used to transfect human hepatoma cell line HepG2.2.15 by calcium phosphate method and liposome method respectively, The expression of HBsAg and HBeAg in the cell culture medium was detected by RIA method 3 days after transfection to observe the inhibitory effect of endogenous antisense RNA on HBV gene expression. The study confirmed that liposome transfection efficiency is higher, indicating that liposomes is the ideal way to carry gene therapy.
