目的探讨动物致癌性实验中肿瘤发生率的统计分析方法。方法通过对传统的Cochran-Armitage趋势检验进行校正,采用Poly-k趋势检验比较药物的不同剂量组动物肿瘤发生率的差异,结合实例给出实现该分析过程的SAS程序。结果对大鼠致癌性实验数据的分析表明,Cochran-Armitage线性趋势检验与Poly-k趋势检验的结果存在一定差异,其各自所反映的肿瘤发生率的趋势不尽相同。结论在致癌性实验中,由于各剂量组生存情况的不同,可能会对肿瘤发生率的分析造成影响,Poly-k趋势检验可有效地解决这一问题。
Objective To investigate the method of statistical analysis of tumor incidence in animal carcinogenicity experiment. Methods The traditional Cochran-Armitage trend test was calibrated. Poly-k trend test was used to compare the tumor incidence rates of different dose groups of animals. The SAS program to achieve this analysis was given with examples. Results The analysis of experimental data of carcinogenicity in rats showed that the results of the Cochran-Armitage linear trend test and the Poly-k trend test showed some differences, and the trends of the incidence of tumors reflected by each of them differed. Conclusion In the carcinogenicity experiment, due to the different survival conditions of each dose group, it may affect the analysis of tumor incidence. Poly-k trend test can effectively solve this problem.
