目的观察17-β-雌二醇对人脐静脉内皮细胞纤溶因子活性、基因表达以及胞内钙离子渡度([Ca(2+)]i)的影响。方法应用发色底物法、细胞原位杂交技术和激光共聚焦扫描显微镜。结果组织型纤溶酶原激活物活性及其基因表达随17-β-雌二醇浓度的增加而呈剂量依赖性增加;单个内皮细胞胞内钙离子浓度在高浓度雌二醇作用下先上升后下降,生理浓度时降低。结论雌激素可以提高纤溶能力、拮抗胞内Ca(2+)浓度升高,为雌激素保护心血管作用机制研究提供了部分实验依据。
Objective To observe the effect of 17-β-estradiol on the activity of fibrinolytic factor, gene expression and [Ca (2 +)] i in human umbilical vein endothelial cells. Methods The chromogenic substrate method, in situ hybridization and laser confocal scanning microscopy were used. Results Tissue-type plasminogen activator and its gene expression increased in a dose-dependent manner with the increase of 17-β-estradiol concentration. The intracellular calcium concentration of single endothelial cells increased first under the action of high concentration of estradiol After falling, physiological concentration decreased. Conclusion Estrogen can increase the ability of fibrinolysis and antagonize the increase of intracellular Ca (2+) concentration, which provides some experimental evidences for the mechanism of estrogen in protecting cardiovascular system.
