建立了分离测定槐角提取液中染料木素的在线酸坝富集-毛细管区带电泳方法,考察了酸的种类、酸的及浓度、进样/进酸时间比例等因素对堆积效率的影响。实验以20 mmol/L硼砂(pH10.5)作为缓冲溶液,200 mmol/L柠檬酸溶液(pH1.7)作为酸坝,进样、进酸时间分别为180s和45s,在分离电压15kV,检测波长254 nm的条件下,染料木素5 min内出峰,检出限可达10 nmol/L,富集倍数为300多倍。 An online acid-dam enrichment-capillary zone electrophoresis method was developed for the separation and determination of genistein in the extract of Sophora japonica. The effects of acid species, acid concentration and injection / acid addition time on the deposition efficiency were investigated . In the experiment, 20 mmol / L borax (pH10.5) was used as the buffer solution and 200 mmol / L citric acid solution (pH1.7) as the acid dams. The time of entering the acid was 180s and 45s respectively. Under the condition of 254 nm wavelength, Genistein peaked within 5 min, the detection limit was up to 10 nmol / L and the enrichment factor was 300 times.
